Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.3418T>G (p.Cys1140Gly), citing Ambry Variant Classification Scheme 2023: The p.C1140G variant (also known as c.3418T>G), located in coding exon 30 of the ANK2 gene, results from a T to G substitution at nucleotide position 3418. The cysteine at codon 1140 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
