Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.3400A>C (p.Lys1134Gln), citing Ambry Variant Classification Scheme 2023: The p.K1134Q variant (also known as c.3400A>C), located in coding exon 22 of the RAD50 gene, results from an A to C substitution at nucleotide position 3400. The lysine at codon 1134 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
