NM_000039.3(APOA1):c.339T>G (p.Asp113Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D113E variant (also known as c.339T>G), located in coding exon 3 of the APOA1 gene, results from a T to G substitution at nucleotide position 339. The aspartic acid at codon 113 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, glutamic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
