Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3386G>A (p.Cys1129Tyr), citing Ambry Variant Classification Scheme 2023: The p.C1129Y variant (also known as c.3386G>A), located in coding exon 5 of the MSH6 gene, results from a G to A substitution at nucleotide position 3386. The cysteine at codon 1129 is replaced by tyrosine, an amino acid with highly dissimilar properties. This variant has been reported in an individual affected with colorectal cancer (Yurgelun MB et al. J Clin Oncol, 2017 Apr;35:1086-1095). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
