NM_001008212.2(OPTN):c.113A>G (p.Glu38Gly) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E38G variant (also known as c.113A>G), located in coding exon 1 of the OPTN gene, results from an A to G substitution at nucleotide position 113. The glutamic acid at codon 38 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
