Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003238.6(TGFB2):c.336C>G (p.Phe112Leu), citing Ambry Variant Classification Scheme 2023: The p.F112L variant (also known as c.336C>G), located in coding exon 1 of the TGFB2 gene, results from a C to G substitution at nucleotide position 336. The phenylalanine at codon 112 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
