NM_000038.6(APC):c.3367C>G (p.Gln1123Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1123E variant (also known as c.3367C>G), located in coding exon 15 of the APC gene, results from a C to G substitution at nucleotide position 3367. The glutamine at codon 1123 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved through mammals but not in all available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
