Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3335A>C (p.Asp1112Ala), citing Ambry Variant Classification Scheme 2023: The p.D1112A variant (also known as c.3335A>C), located in coding exon 5 of the MSH6 gene, results from an A to C substitution at nucleotide position 3335. The aspartic acid at codon 1112 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
