Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3470A>G (p.Asn1157Ser), citing Ambry Variant Classification Scheme 2023: The p.N1111S variant (also known as c.3332A>G), located in coding exon 29 of the KIF1B gene, results from an A to G substitution at nucleotide position 3332. The asparagine at codon 1111 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
