NM_001267550.2(TTN):c.60202G>C (p.Glu20068Gln) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E11003Q variant (also known as c.33007G>C), located in coding exon 130 of the TTN gene, results from a G to C substitution at nucleotide position 33007. The glutamic acid at codon 11003 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
