Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.32G>C (p.Cys11Ser), citing Ambry Variant Classification Scheme 2023: The p.C11S variant (also known as c.32G>C), located in coding exon 1 of the ATM gene, results from a G to C substitution at nucleotide position 32. The cysteine at codon 11 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
