Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.325A>G (p.Arg109Gly), citing Ambry Variant Classification Scheme 2023: The p.R109G variant (also known as c.325A>G), located in coding exon 4 of the POLE gene, results from an A to G substitution at nucleotide position 325. The arginine at codon 109 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
