Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.324T>G (p.Cys108Trp), citing Ambry Variant Classification Scheme 2023: The p.C108W variant (also known as c.324T>G), located in coding exon 2 of the CHEK2 gene, results from a T to G substitution at nucleotide position 324. The cysteine at codon 108 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
