Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.3230C>G (p.Ser1077Cys), citing Ambry Variant Classification Scheme 2023: The p.S1077C variant (also known as c.3230C>G), located in coding exon 8 of the AKAP9 gene, results from a C to G substitution at nucleotide position 3230. The serine at codon 1077 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
