Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.3167G>A (p.Ser1056Asn), citing Ambry Variant Classification Scheme 2023: The p.S1074N variant (also known as c.3221G>A), located in coding exon 14 of the MET gene, results from a G to A substitution at nucleotide position 3221. The serine at codon 1074 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
