NM_004281.4(BAG3):c.321G>C (p.Gln107His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q107H variant (also known as c.321G>C), located in coding exon 2 of the BAG3 gene, results from a G to C substitution at nucleotide position 321. The glutamine at codon 107 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
