NM_001110556.2(FLNA):c.3212A>C (p.Lys1071Thr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1071T variant (also known as c.3212A>C), located in coding exon 21 of the FLNA gene, results from an A to C substitution at nucleotide position 3212. The lysine at codon 1071 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
