Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000335.5(SCN5A):c.3211G>A (p.Glu1071Lys), citing Ambry Variant Classification Scheme 2023: The p.E1071K variant (also known as c.3211G>A), located in coding exon 16 of the SCN5A gene, results from a G to A substitution at nucleotide position 3211. The glutamic acid at codon 1071 is replaced by lysine, an amino acid with similar properties, and is located in the interdomain linker DII/DIII region. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
