NM_003036.4(SKI):c.320G>A (p.Gly107Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G107D variant (also known as c.320G>A), located in coding exon 1 of the SKI gene, results from a G to A substitution at nucleotide position 320. The glycine at codon 107 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
