NM_000393.5(COL5A2):c.3200G>C (p.Arg1067Pro) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1067P variant (also known as c.3200G>C), located in coding exon 45 of the COL5A2 gene, results from a G to C substitution at nucleotide position 3200. The arginine at codon 1067 is replaced by proline, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
