Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.3472C>T (p.His1158Tyr), citing Ambry Variant Classification Scheme 2023: The p.H1066Y variant (also known as c.3196C>T), located in coding exon 9 of the OBSCN gene, results from a C to T substitution at nucleotide position 3196. The histidine at codon 1066 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
