Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001134363.3(RBM20):c.3190G>T (p.Asp1064Tyr), citing Ambry Variant Classification Scheme 2023: The p.D1064Y variant (also known as c.3190G>T), located in coding exon 11 of the RBM20 gene, results from a G to T substitution at nucleotide position 3190. The aspartic acid at codon 1064 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
