NM_001184.4(ATR):c.1010T>C (p.Leu337Pro) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L337P variant (also known as c.1010T>C), located in coding exon 4 of the ATR gene, results from a T to C substitution at nucleotide position 1010. The leucine at codon 337 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
