NM_004387.4(NKX2-5):c.317C>G (p.Pro106Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P106R variant (also known as c.317C>G), located in coding exon 1 of the NKX2-5 gene, results from a C to G substitution at nucleotide position 317. The proline at codon 106 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
