NM_000245.4(MET):c.3119C>G (p.Ser1040Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1058C variant (also known as c.3173C>G), located in coding exon 14 of the MET gene, results from a C to G substitution at nucleotide position 3173. The serine at codon 1058 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
