Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.3148G>A (p.Gly1050Ser), citing Ambry Variant Classification Scheme 2023: The p.G1050S variant (also known as c.3148G>A), located in coding exon 19 of the NOTCH1 gene, results from a G to A substitution at nucleotide position 3148. The glycine at codon 1050 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
