NM_000179.3(MSH6):c.3144G>C (p.Gln1048His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1048H variant (also known as c.3144G>C), located in coding exon 4 of the MSH6 gene, results from a G to C substitution at nucleotide position 3144. The glutamine at codon 1048 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
