NM_001148.6(ANK2):c.11308C>A (p.Gln3770Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q3770K variant (also known as c.11308C>A), located in coding exon 42 of the ANK2 gene, results from a C to A substitution at nucleotide position 11308. The glutamine at codon 3770 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, lysine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
