Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.3141G>C (p.Glu1047Asp), citing Ambry Variant Classification Scheme 2023: The p.E1047D variant (also known as c.3141G>C), located in coding exon 16 of the DNAH11 gene, results from a G to C substitution at nucleotide position 3141. The glutamic acid at codon 1047 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
