NM_000722.4(CACNA2D1):c.3140_3157dup (p.Val1052_Leu1053insCysPheAspAsnAsnVal) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the CACNA2D1 gene (transcript NM_000722.4) at coding-DNA position 3140 through coding-DNA position 3157, duplicating 18 bases. Submitter rationale: The c.3140_3157dup18 variant (also known as p.C1047_V1052dup), located in coding exon 38 of the CACNA2D1 gene, results from an in-frame duplication of 18 nucleotides at nucleotide positions 3140 to 3157. This results in the duplication of 6 extra residues (CFDNNV) between codons 1047 and 1052. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
