Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.3115G>A (p.Gly1039Arg), citing Ambry Variant Classification Scheme 2023: The p.G1039R variant (also known as c.3115G>A), located in coding exon 23 of the BUB1B gene, results from a G to A substitution at nucleotide position 3115. The glycine at codon 1039 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
