NM_002691.4(POLD1):c.3114G>C (p.Gln1038His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1038H variant (also known as c.3114G>C), located in coding exon 24 of the POLD1 gene, results from a G to C substitution at nucleotide position 3114. The glutamine at codon 1038 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
