Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004006.3(DMD):c.310C>T (p.His104Tyr), citing Ambry Variant Classification Scheme 2023: The p.H104Y variant (also known as c.310C>T), located in coding exon 5 of the DMD gene, results from a C to T substitution at nucleotide position 310. The histidine at codon 104 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
