NM_000371.4(TTR):c.308G>A (p.Gly103Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G103D variant (also known as c.308G>A), located in coding exon 3 of the TTR gene, results from a G to A substitution at nucleotide position 308. The glycine at codon 103 is replaced by aspartic acid, an amino acid with similar properties. Another alteration at this position (p.G103R also known as p.G83R in legacy nomenclature) has been reported in the literature in association with familial amyloid polyneuropathy (Liu T et al. Eye (Lond), 2014 Jan;28:26-33). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for the p.G103D alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
