Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002519.3(NPAT):c.3088A>C (p.Lys1030Gln), citing Invitae Variant Classification Sherloc (09022015): An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This sequence change replaces lysine, which is basic and polar, with glutamine, which is neutral and polar, at codon 1030 of the NPAT protein (p.Lys1030Gln). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NPAT-related conditions. ClinVar contains an entry for this variant (Variation ID: 1727412). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
