NM_001252024.2(TRPM1):c.4013T>G (p.Leu1338Arg) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TRPM1 c.3947T>G (p.Leu1316Arg) results in a non-conservative amino acid change located in the Ion transport domain (IPR005821) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 249548 control chromosomes (gnomAD). c.3947T>G has been reported in the literature in at-least one homozygous individual affected with Congenital Stationary Night Blindness (example: Habib_2020). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
