Likely pathogenic for Bardet-Biedl syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000007.13:g.(33427757_33545074)_(33573789_33644476)del, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exons 20-21 in the BBS9 gene. A presumed nomenclature of c.(2115+1_2116-1)_(2521+1_2522-1)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a frameshift in the BBS9 gene, a known mechanism of disease. The variant was absent in 21694 control chromosomes (gnomAD, Structural Variants dataset). To our knowledge, no occurrence of c.(2115+1_2116-1)_(2521+1_2522-1)del in individuals affected with Bardet-Biedl Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. A ClinVar submitter (evaluation after 2014) cites the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as likely pathogenic.
