This variant lies in the PEX7 gene (transcript NM_000288.4) at coding-DNA position 401, where A is replaced by T; at the protein level this means replaces aspartic acid at residue 134 with valine — a missense variant. Submitter rationale: Variant summary: PEX7 c.401A>T (p.Asp134Val) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251418 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.401A>T has been reported in the literature together with a pathogenic variant in an individual affected with Rhizomelic Chondrodysplasia Punctata Type 1, however it was not specified whether the variants were in cis or in trans (e.g. Duker_2016).
