Pathogenic for Anemia, nonspherocytic hemolytic, due to G6PD deficiency — the classification assigned by Dunham Lab, University of Washington to NM_001360016.2(G6PD):c.496C>T (p.Arg166Cys), citing ACMG Guidelines, 2015: Variant reported in hemizygote with G6PD deficiency and acute anemia after exposure to oxidative stressor, and in another unrelated hemizygote with recurrent infections and altered response to ROS (PP4, PS4_M). Undetectable activity reported in red blood cells of hemizygote (PS3). Not observed in gnomAD (PM2). In one report, mother of hemizygote does not have variant, so presumed de novo (PM6). In another, hemizygote and heterozygous mother both have variant and altered response to ROS (PP1). Predicted to alter secondary structure by disrupting local hydrophobic interactions by introduction of additional cysteine (PP3). Post_P 0.999 (odds of pathogenicity 13661, Prior_P 0.1).
