Likely pathogenic for Infantile epileptic dyskinetic encephalopathy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004341.5(CAD):c.2378_2379del (p.Thr793fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: CAD c.2378_2379delCA (p.Thr793SerfsX15) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncating variants downstream of this position have been reported in association with CAD deficiency and Epileptic encephalopathy in HGMD. The variant was absent in 251274 control chromosomes (gnomAD). To our knowledge, no occurrence of c.2378_2379delCA in individuals affected with Early Infantile Epileptic Encephalopathy, 50 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
