NM_000059.4(BRCA2):c.5713C>T (p.His1905Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1905Y variant (also known as c.5713C>T), located in coding exon 10 of the BRCA2 gene, results from a C to T substitution at nucleotide position 5713. The histidine at codon 1905 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
