Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.1273T>C (p.Ser425Pro), citing Ambry Variant Classification Scheme 2023: The p.S425P variant (also known as c.1273T>C), located in coding exon 9 of the BRCA1 gene, results from a T to C substitution at nucleotide position 1273. The serine at codon 425 is replaced by proline, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
