Uncertain significance for Ullrich congenital muscular dystrophy 2; Bethlem myopathy 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004370.6(COL12A1):c.7360A>C (p.Ser2454Arg), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with COL12A1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 2454 of the COL12A1 protein (p.Ser2454Arg).
