Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.870C>G (p.Phe290Leu), citing Ambry Variant Classification Scheme 2023: The p.F290L variant (also known as c.870C>G), located in coding exon 8 of the PMS2 gene, results from a C to G substitution at nucleotide position 870. The phenylalanine at codon 290 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
