Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.3216C>A (p.Asp1072Glu), citing Ambry Variant Classification Scheme 2023: The p.D1072E variant (also known as c.3216C>A), located in coding exon 27 of the EGFR gene, results from a C to A substitution at nucleotide position 3216. The aspartic acid at codon 1072 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
