This variant lies in the MYO7A gene (transcript NM_000260.4) at coding-DNA position 5994, where G is replaced by T; at the protein level this means replaces tryptophan at residue 1998 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tryptophan, which is neutral and slightly polar, with cysteine, which is neutral and slightly polar, at codon 1998 of the MYO7A protein (p.Trp1998Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive deafness and/or clinical features of Usher syndrome (PMID: 36164746; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1719700).
