Uncertain significance for T-cell immunodeficiency with epidermodysplasia verruciformis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004310.5(RHOH):c.310A>G (p.Ser104Gly), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs765575265, gnomAD 0.02%). This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 104 of the RHOH protein (p.Ser104Gly). This variant has not been reported in the literature in individuals affected with RHOH-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
