Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.2270A>G (p.His757Arg), citing Ambry Variant Classification Scheme 2023: The p.H757R variant (also known as c.2270A>G), located in coding exon 9 of the AXIN2 gene, results from an A to G substitution at nucleotide position 2270. The histidine at codon 757 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
