In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Asp113 amino acid residue in SDHD. Other variant(s) that disrupt this residue have been observed in individuals with SDHD-related conditions (PMID: 34906457; Invitae), which suggests that this may be a clinically significant amino acid residue. This missense change has been observed in individual(s) with clinical features of autosomal dominant hereditary paraganglioma-pheochromocytoma (PGL-PCC) syndrome (Invitae). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 113 of the SDHD protein (p.Asp113His).
