Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.2571A>C (p.Leu857Phe), citing Ambry Variant Classification Scheme 2023: The p.L857F variant (also known as c.2571A>C), located in coding exon 23 of the POLE gene, results from an A to C substitution at nucleotide position 2571. The leucine at codon 857 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
